Summary:PURPOSE: Provided are methods of culture, extraction and analysis for producing an active anti-tumor substance, decursinol angelate, via a root culture of Angelica gigas Nakai, thereby stably mass-producing decursinol angelate in a short time, regardless of environmental conditions, such as weather, soils, and damage from disease and harmful insects. CONSTITUTION: Angelica gigas Nakai. roots are aseptic-cultured in a flask containing 3-indolebutyric acid as a plant growth regulator and methyl jasmonate as an elicitor. The active anti-tumor substance, decursinol angelate, represented by the formula(1) is extracted by the steps of: 1) adding ethanol to the cultured root tissue and heating it at 50deg.C.; 2) evaporating ethanol from the resulting extract solution; 3) adding the same amount of chloroform to the resulting extract and extracting decursinol into an organic solvent layer; 4) obtaining the residue after evaporating chloroform. The decursinol angelate is analyzed by high performance liquid chromatography(HPLC) at 330nm using a reverse phase C-18 column.
