Abstract PURPOSE: Novel inhibitors of matrix metalloproteinases (MMPs) are provided which is useful for the prevention, control and treatment of diseases in which the proteolytic action of MMPs are involved. CONSTITUTION: Succinic amide derivatives of formula (I), wherein W is a -CO2H or-CONHOH group; R, R1 and R2 are each hydrogen or an organic residue, R3 is the residue of an alpha-amino acid and R4 is an organic group, are inhibitors of matrix metalloproteinases (MMPs) and inhibit the release of tumor necrosis factor-alpha (TNF) from cells. Therefore the derivatives are useful for the prevention, control and treatment of diseases in which MMPs or TNF are involved. 화학식 I의 숙신산 아미드 유도체는 매트릭스 메탈로프로티나아제(MMP)의 억제제로서, 세포로부터 종양 괴저 인자-알파(TNF)가 방출되는 것을 억제시키므로 MMP 또는 TNF가 관련되는 질환을 예방, 조절 및 치료하는데 유용하다. 본원에는 또한 당해 화합물의 제조방법 및 이를 함유하는 약제학적 조성물이 기술되어 있다. 화학식 I 상기식에서, W는 -COH 또는 -CONHOH 그룹이고, R, R및 R는 각각 수소 또는 유기 잔기이며, R은 알파-아미노산의 잔기이고, R는 유기 그룹이다.
