PURPOSE: Provided are variants of parathyroid hormone(PTH), a process for their production, pharmaceutical preparations comprising them and their use as a pharmaceutical. CONSTITUTION: A PTH compound has PTH-like activity and comprises at least one modification, wherein the modification is either (1) at least one radical selected from an L- or D-alpha-amino acid,C2-6 alkoxycarbonyl and optionally substituted C1-8 alkyl, C2 -8 alkenyl, C2-8 alkynyl, aralkyl, aralkenyl or C3-6 cycloalkyl-C1-4 alkyl and attached to the terminal amino group of the PTH compound, and/or at least one radical selected from C26 alkoxycarbonyl and optionally substituted C1-8 alkyl, C2-8 alkenyl, C2-8 alkynyl, aralkyl, aralkenyl or C3-6 cycloalkyl-C1-4 alkyl and attached to one or more side chain amino groups of the PTH compound, or (2) at least one a-amino acid unit in the positions 1 to 38 of a naturally occurring PTH sequence being replaced by a natural or unnatural amino acid unit optionally in protected form, whereby the a-amino acid units present in positions 1 and 2 at the amino terminus of the PTH sequence together are replaced by a pseudo-peptide, or a combination of such modifications. It has pharmacological activity, for example, for preventing or treating all bone conditions which are associated with increased calcium depletion or resorption or in which calcium fixation in the bone is desirable. 본 발명은 PTH-유사 활성을 가지며 하나 이상의 수사(修篩)를 함유하는 유리 헝태, 염 형태의 PTH화합물에 관한 것으로서, 상기 수사는 1. 하나이상의 라디칼이 L-또는 D-α-아미노산, C알콕시카르보닐 및 임의 치환된 C알킬, C알케닐, C알키닐, 아르알킬, 아르알케닐 또는 C시클로알킬-C알킬 중에서 선택되고 상기 PTH 화합물의 말단 아미노기에 결합된 것이나, 및/또는 하나이상의 라디칼이 C알콕시카르보닐 및 임의 치한된 C알킬, C알케닐, C알키닐, 아르알킬, 아르알케닐 또는 C시클로알킬-C알킬 중에서 선택되고 상기 PTH 화합물의 하나 이상의 측쇄 아미노기에 결합된 것이거나, 또는 2. 자연 발생적인 PTH 서열의 1 내지 38 위치내에 하나이상의-아미노산 유니트가 임의의 보호 헝태의 천연 또는 비천연적 아미노산 유니트로 대체되어 이로 인해 상기 PTH 서열의 아미노 말단의 위치 1 및 2에 존재하는-아미노산 유니트들이 가성(假性)-펩티드로 대체될 수 있는 것이거나, 또는 그런 수사들의 복합물일 수도 있으며, 그러한 상기 PTH화합물은 예를들면 칼슘 고갈이나 재흡수의 증가와 관련되거나 또는 골격내에 칼슘 고정이 필요로 되는 모든 골격이상을 예방 또는 치료하기위한 약리학적 활성을 가지고 있다.
