Summary:PURPOSE: To obtain a bovine Y chromosome specific DNA and/or a marker containing a DNA common to male and female useful for distinguishing a sex chromosome from an autosomal chromosome, an X CHR from a Y CHR and a short arm from a long arm in the Y CHR. CONSTITUTION: A female bovine DNA is isolated, a fragment having no regularity in DNA sequences at both ends is prepared from the DNA, is mixed with a DNA fragment obtained by treating a male bovine DNA with II type restriction enzyme and preparing regular ends and reassociated. Only a duplex DNA having regular ends is incorporated into a cloning vector having a restriction end contemporary with the ends, transduced into a host to give a DNA fragment having a male specific sequence, its base sequence is determined to give a sequence of the formula. The DNA fragment is used as a probe, a second and a third DNAs having male specific sequences are isolated from a male bovine DNA genome library, and a fourth DNA providing different association images between male and female and a fifth to a seventh DNAs having sequences common to male and female are obtained by Southern blot method.
