Summary:PURPOSE:To obtain the subject novel DNA fragment capable of strongly expressing a useful gene in a coryne type bacterium and capable of controlling the expression of the gene by changing the compositional condition of a culture medium, etc. CONSTITUTION:This DNA fragment is obtained from the chromosome of a coryne type bacterium such as Brevibacterium flavum MJ-233, has a stronger function than of a tac promoter in the coryne tyep bacterium, and has e.g. a base sequence of the formula. The DNA fragment is obtained from the coryne bacterium with a vector used for the detection of plasmids and having (a) a plasmid-replicating and multiplying DNA region functioning in Escherichia coil, (b) a plasmid-replicating and multiplying DNA region functioning in the coryne type bacterium, (c) a DNA region containing a plasmid marker gene, (d) a DNA region containing a reporter gene, and (e) a transcription termination factor placed on the upstream of the (d) region.
