Summary:PURPOSE:To enable fixing of a specimen in a natural state without causing a chemical change of the tissue of the specimen, by sealing the specimen and a physiological saline solution in a bag-shaped flexible vessel made of a resin film, by charging this flexible vessel in a high-pressure vessel filled up with a pressure medium liquid and by pressurizing the specimen isotropically through the intermediary of the pressure medium liquid. CONSTITUTION:A construction is made of a high-pressure vessel 1 having a pressurizing process chamber 3 in which a specimen is put, a top cap 2, a piston 4 for pressurization, a hydraulic cylinder 5 and a hydraulic unit 6. A specimen A is charged in the high-pressure vessel 1, a pressure medium 10 such as a physiological saline solution is filled up around it and thereafter the pressurizing process chamber 3 is closed hermetically by the top cap 2. Next, the piston 4 is made to rise by operating the hydraulic unit 6. Since the compressibility of the pressure medium liquid 10 is very small, then, a liquid pressure in the process chamber 3 rises rapidly and the specimen A is pressurized isotropically from all directions. When the pressure reaches a pressure of fixing protein constituting the specimen A, the fixing the specimen A starts rapidly and the fixing is completed in a short time.
