Summary:PURPOSE:To establish the method for measurement with high accuracy by optically measuring a rheumatoid factor by using a reagent formed by incorporating chemically modified human gamma globulin therein. CONSTITUTION:The rheumatoid factor is optically measured by using the reagent formed by incorporating the chemically modified human gamma globulin therein. The method for chemically modifying the human gamma globulin is exemplified by an acid anhydride method using alkylchloroformate in addition to glutaraldehyde, etc., and a protein crosslinking agent such as carbodiimide and maleimide. Among these method, the glutaraldehyde treatment is more preferable in terms of ease and in terms of the efficient formation of self- polymn. The unreacted glutaraldehyde is neutralized by adding monoethanol amine thereto and is removed by dialysis with a large volume of 0.5% NaCl water as the simpler method.
