Abstract PROBLEM TO BE SOLVED: To provide pluripotent stem cells from non embryonic cells or postnatal animal cells or tissue, capable of differentiating not limited by the lineage of the source cells or tissue.SOLUTION: The invention provides a method for isolating from non embryonic cells or cells or tissues of a postnatal animal a pluripotent embryonic-like stem cell capable of self regeneration and capable of differentiating into cells of endodermal, ectodermal and mesodermal lineages and making the cells into a differentiable state, comprising the steps of: (a) obtaining cells from a postnatal animal source; (b) slowly freezing the cells in a culture medium containing 7.5%(v/v) dimethylsulfoxide until a final temperature of -80°C is reached; and (c) culturing the cells.SELECTED DRAWING: None 【課題】 非胚細胞もしくは生後動物細胞または組織から多能性幹細胞であって、単離された細胞もしくは組織の系統に制限されず分化できる多能性幹細胞を提供すること。【解決手段】 (a)生後動物供給源から細胞を入手する工程と、(b)-80℃の最終温度に到達するまで、7.5%(v/v)ジメチルスルホキシドを含有する培地中で前記細胞を緩徐に凍結する工程と、(c)前記細胞を培養する工程と、を含む、自己再生可能ならびに内胚葉、外胚葉および中胚葉系統の細胞に分化可能な多能性胚様幹細胞を非胚細胞もしくは生後動物細胞または組織から単離し、分化可能な状態にする。【選択図】 なし
