Abstract | PROBLEM TO BE SOLVED: To provide a method of refining interleukin-4 or related variant from Escherichia coli culture media where interleukin-4 and its derivatives are developed as an insoluble protein aggregate, so-called inclusion body. SOLUTION: This method of preparation of mutated protein of interleukin-4 comprises (a) development in an inclusion body, (b) separation of the inclusion body by decomposition of a cell, (c) cleaning of the inclusion body thus obtained, (d) solubilization of the inclusion body by denaturation, (e) restoration of the developed product and (f) refining of the developed product. In (c) the inclusion body is cleaned by a buffer containing a detergent which effectively solubilizes fat bonded on the surface of the inclusion body or contained in cell wall scraps and restoration (e) is carried out in the presence of an artificial chaperon.
