Abstract | The invention provides a shRNA (short hairpin Ribonucleic Acid) adeno-associated virus vector as well as a construction method and application thereof, and belongs to the technical field of biological medicines. The invention provides shRNA for targeted inhibition of ARG1 expression, a shuttle plasmid, a recombinant adeno-associated virus vector and a recombinant adeno-associated virus are constructed based on the shRNA, the recombinant adeno-associated virus (AAV) is used as the vector and carries an ARG1-shRNA sequence, and the ARG1-shRNA interference plasmid can significantly and specifically inhibit the ARG1 expression, so that the ARG1 expression can be inhibited, and the ARG1 expression can be inhibited. The polypeptide has the effect of resisting vascular endothelial dysfunction induced by the type 2 diabetes, is high in safety and provides a novel ARG1 targeting treatment strategy for cardiovascular diseases of the type 2 diabetes.
