Abstract | The invention discloses a nano preparation as well as a preparation method and application thereof, and relates to the technical field of medical materials. The nanoformulations of the present invention comprise a first polymer grafted with norcantharidin and/or a second polymer grafted with an antioxidant. The nano preparation provided by the invention can realize weak acid response release of norcantharidin in a tumor microenvironment, so that inhibitory immune cell differentiation is regulated and controlled, inhibitory escape of tumors to immune response is weakened, active oxygen generated by radiotherapy in normal cells can be eliminated, oxidative damage is avoided, and side effects of radiotherapy are reduced. In addition, by removing active oxygen, the stimulation activity of an injury-related molecular mode on dendritic cells can be recovered, the killing function of infiltrated T cells in a tumor microenvironment can be recovered, and adaptive immunity caused by radiotherapy can be enhanced.
