Abstract The invention belongs to the field of biomedical engineering, and provides a novel recombinant protein which comprises a fusion protein formed by a TMEM8B-a protein and a reporter protein, and a novel proteasome inhibitor can be screened with high throughput by using the recombinant protein. The invention also correspondingly provides a high-throughput screening method of the proteasome inhibitor. The screening method is expected to quickly and effectively screen a new generation of proteasome inhibitor medicine which is better in effect, stronger in specificity, smaller in side effect and also plays a role in solid tumors. And for the high-throughput screening method, the whole process can be completed in a living cell state, and cell lysis or addition of an additional color development or luminescence reaction reagent is not needed, so that the high-throughput screening method is simple, convenient, rapid and accurate.
