Summary: | The invention discloses a non-natural base pair formed by specific pairing of m3T and TPT3 as well as a derivative and application of the non-natural base pair, and the structural general formula of the non-natural base pair is shown in the specification. N3 methylated thymine in DNA is taken as a target, and bases capable of being specifically paired with N3 methylated thymine are screened by utilizing a kinetic method. And the screened TPT3 can be specifically paired with N3 methyl thymine. After the TPT3 and the m3T are specifically paired, the DNA can be effectively extended and amplified. The m3T-TPT3 can be converted into a NaM-TPT3 non-natural base pair in the amplification process, and the N3 methylated thymine site can be judged by comparing the termination of a sanger sequencing signal. The m3T-TPT3 non-natural base pair can recognize and capture N3 alkylation damage in complex biological samples such as low-abundance N3 methylated thymine and plasmids, and has great potential in application to DNA |
