Abstract The invention discloses a beta-glucosidase mutant, an engineering bacterium and application of the beta-glucosidase mutant in degradation of cellulose. The beta-glucosidase mutant is obtained by performing site-specific mutagenesis on the 20th site or the 240th site of an amino acid sequence as shown in SEQ ID No.2. The specific activity of the mutant enzyme provided by the invention is obviously improved by 1.5 times or more than that of a wild enzyme. When a mixed solution of endoglucanase, exoglucanase, lysed polysaccharide monooxygenase and mutant enzyme is used as a catalyst, the yield of glucose is increased by 15-25% compared with that of wild enzyme. 本发明公开了一种β-葡萄糖苷酶突变体、工程菌及其在降解纤维素中的应用,所述β-葡萄糖苷酶突变体是将SEQ ID No.2所示氨基酸序列第20位或第240位进行定点突变获得的。本发明突变酶的比活力比野生酶显著提高1.5倍及以上。以内切葡聚糖苷酶、外切葡聚糖苷酶、裂解多糖单加氧酶与突变酶的混合液为催化剂时,葡萄糖产率较野生酶提高了15%-25%。
AbstractList The invention discloses a beta-glucosidase mutant, an engineering bacterium and application of the beta-glucosidase mutant in degradation of cellulose. The beta-glucosidase mutant is obtained by performing site-specific mutagenesis on the 20th site or the 240th site of an amino acid sequence as shown in SEQ ID No.2. The specific activity of the mutant enzyme provided by the invention is obviously improved by 1.5 times or more than that of a wild enzyme.
