Abstract The invention discloses a method for purity identification of watermelon hybrid seeds based on an SSR marker method. The method comprises: taking the genomic DNA of a single plant of a hybrid watermelon leaf as a template, adding upstream primers and downstream primers, carrying out PCR amplification, and obtaining an amplified product; gel electrophoresis is carry out on that amplification product, EB staining is carry out, and imaging is carried out in a gel imaging system; according to the imaging results, the band pattern was counted. The purity of watermelon hybrid seeds was calculated according to the results of banding pattern. Molecular marker technology directly reveals the differences between parents at DNA level, so the results are accurate and reliable, and can be identified atthe seedling stage or even seed level, so the experimental cycle is greatly shortened.
