The invention provides a preparation method of an enteromorpha polysaccharide microsphere. The preparation method comprises the following steps: (A) mixing paraffin oil with Span 80, so as to obtain acontinuous phase of water in oil type emulsified liquid; (B) adding an enteromorpha polysaccharide water solution into the continuous phase, mixing, and stirring, so as to obtain the water in oil type emulsified liquid; and (C) sequentially carrying out low-temperature solidification and centrifugation on the water in oil type emulsified liquid, and removing supernatant liquid, so as to obtain the enteromorpha polysaccharide microsphere. By preparing the enteromorpha polysaccharide microsphere by virtue of an emulsifying method, the preparation method is simple and relatively good in repeatability; and no solidifying agent needs to be added in the preparation process, so that the prepared enteromorpha polysaccharide microsphere is natural and toxic-free and relatively good in dispersity and spherical degree, and
