Summary: | The present invention belongs to the technical field of biology, and particularly relates to a method for promoting protein N-terminal identification by using laser-assisted enzymolysis. The method comprises: carrying out amino blocking on a protein, and adding trypsin (2.5-5% of the amount of the protein); vertically irradiating the blocked protein for 5-80 s with 808 nm infrared laser so as to carry out laser-assisted enzymolysis; and finally identifying the protein N-terminal peptide fragment by using matrix assisted laser desorption ionization time of flight mass spectrum or liquid chromatograph mass spectrometer. With the method of the present invention, the enzymolysis time required by the protein N-terminal peptide fragment identification can be substantially shortened, the N-terminal peptide fragment signal of the single protein is enhanced compared to the conventional solution enzymolysis, and the N-terminal peptide fragment identification number of the complex proteome sample can be improved. |
