The invention discloses an extracting and separating method for various active ingredients of coix seeds. The extracting and separating method includes the steps that after the coix seeds are smashed, prilled and dried, coix seed ester, coix seed oil and degreased coix seed particles are extracted with the supercritical CO2 extraction technology, wherein the yield of the coix seed ester is 0.54%, and the purity of the coix seed ester is 89.97%; assisted aqueous extracting is carried out on the degreased coix seed particles through thermostable amylase, coix seed starch is hydrolyzed, and water extract and coix seed proteins are obtained at the same time, wherein the yield of the coix seed proteins is 21.5%, and the purity of the coix seed proteins is 57.70%; enzymolysis is carried out on the water extract through saccharifying enzymes, and enzymatic hydrolysate is obtained; the enzymatic hydrolysate is separated with the membrane separation technology, and retention fluid and filtered fluid are obtained; the retention fluid is dried, and coix seed extract is obtained, wherein the yield of the coix seed extract is 0.92%, the yield of polysaccharide in the coix seed extract is 0.53%, and the purity of the polysaccharide is 57.61%. The obtained coix seed ester, the obtained coix seed oil, the obtained coix seed proteins and the obtained coix seed extract can be directly used as raw materials of common food, health food or special medical application formula food, and the coix seeds are comprehensively used.
