Abstract | The invention belongs to the field of pharmacy, and particularly relates to pegylated melanin nano-particles and a preparation method and application thereof. The preparation method includes the following steps that firstly, melanin is taken and dissolved in an alkaline aqueous solution, acid is added under the stirring condition so that the pH can be adjusted to 7.0, centrifuging is performed, washing is performed, and freezing and drying are performed so that water-soluble melanin nano-particles can be obtained, and the melanin nano-particles are prepared for use; secondly, polyethylene glycol with the ends containing amino groups is used for perform surface treatment on the water-soluble melanin nano-particles. The particle diameter of the pegylated melanin nano-particles prepared through the method is 1-10 nm, the dispersibility of the pegylated melanin nano-particles in water and other solvents is good and can be chelated with Fe3+. An ICR mouse model experiment result shows that the pegylated melanin nano-particles have super-strong iron discharge efficiency and good safety, the half-life period can be as long as 23 h, the drug administration frequency is reduced, the drug administration interval is prolonged, drug use convenience is improved, and the pegylated melanin nano-particles can serve as medicine for treating iron overloaded diseases. |
