Summary: | The invention relates to an in vitro culturing method for parasitic nematodes in the parasitic stage. The method includes the following steps of (1) culture medium preparation and (2) culture. The step (2) includes 1) selecting snails infecting parasitic nematodes, processing the snails infecting the parasitic nematodes and taking the nematodes in the snails to the surface of a flat plate of a solid culture medium; 2) utilizing a pipettor to absorb a solution C to the surface of the flat plate of the solid culture medium; 3) culture the culture medium with nematode bodies at the temperature of 15-22 DEG C; 4) supplementing the solution C every day to keep the liquid level thickness at 0.5cm, opening a cover every day, swinging the culture medium horizontally to enable the nematode bodies to be in full contact with oxygen to obtain the parasitic nematodes after 23 days of culture. The method is simple and convenient to operate. The physiological and biochemical characters of the parasitic nematodes in the parasitic life stage are analyzed. By means of the method, the A. cantonensis L3 (infectious 3-stage larva bodies in the parasitic life stage) in vitro culture technology is built, the parasitic nematodes of the parasitic life stage are cultured in vitro for the first time, and the method provides technical support for scientific research of the parasitic nematodes. |
