The invention relates to a pharmaceutical composition of a blood platelet ADP receptor antagonist and B vitamins. The pharmaceutical composition is formed by the treatment effective dose of the blood platelet ADP receptor antagonist and one of the medical precursor, active metabolites and salt of the blood platelet ADP receptor antagonist, one or more of B-vitamins of the treatment effective dose, and carriers which are acceptable in pharmacy, wherein the blood platelet ADP receptor antagonist comprises ticlopidine, clopidogrel, prasugrel, ticagrelor, elinogrel and the like. The B-vitamins comprise vitamin B6, vitamin B12, folic acid compounds and optimized folic acid compounds. The invention further relates to application of the pharmaceutical composition in preparing drugs for preventing thromboembolic diseases. Drugs made of the pharmaceutical composition have the more remarkable effect of preventing or delaying the thromboembolic diseases on the basis of effectively inhibiting blood platelet functions. In
