Abstract | Post-translational modifications (PTMs) of proteins are known to be altered during breast cancer development. These PTMs are potentially useful biomarkers for breast cancer. In order to study the potential of PTMs in the early detection of breast cancer, ELISA microarray protocols for analyzing several PTMs in a panel of 24 proteins that are found in blood but known to be secreted by breast tissue and/or breast tumors. We evaluated these PTMs in a case-control study with 68 plasma samples. The control samples were from age-matched women with benign breast disease. The sensitivity and the specificity of each assay and the combined assays were evaluated using the areas under (AUC) the receiver operator curves. In the best multivariate analysis, the AUC values ranged from 77% to 87% for 4 different PTMs. Overall, our data suggest that the PTM ELISA microarray platform is a promising tool for discovery and evaluation of biomarkers that have potential for the early detection of breast cancer.
