Abstract 본 연구에서는 피부미용에 효과적인 천연 소재 개발의 일환으로 감마선 조사 기술을 활용하여 rutin의 분해산물의 생성 및 tyrosinase 저해활성 상승을 확인하였다. 우수한 tyrosinase 저해활성 성분의 동정을 위하여 ODS gel을 충진제로 활용한 column chromatography를 수행하여 3종의 rutin 유도체 화합물을 분리하였고, 각 화합물의 구조는 NMR 스펙트럼 분석을 통하여 radiorutinol(1), isoradiorutinol(2) 및 radiorutindiol(3)로 동정하였다. 분리된 화합물에 대해 tyrosinase 저해활성 평가를 수행한 결과, radiorutindiol(3)이 가장 우수한 저해활성(IC50값=45.3±1.6 μM)을 나타내었으며, radiorutinol(1) 및 isoradiorutinol(2)은 각각 71.6±5.9와 56.0±2.1 μM의 IC50 값을 나타냄을 확인하였다. 이들 활성은 rutin의 이중결합 또는 ketone기 위치에 hydroxymethyl화되어 이들 구조 변화가 tyrosinase 저해활성 상승에 영향을 미치는 것으로 추정된다. 향후 이들 활성물질의 활성 기작에 대한 추가적인 연구가 필요하며 tyrosinase 저해활성을 가지는 선도화합물 발굴을 위한 기초자료로 활용이 가능할 것으로 판단된다. Melanin is one of the most important factors affecting skin color, and inhibition of tyrosinase activity can effectively control melanin biosynthesis in the skin. This study was conducted to search and identify potential whitening ingredients from irradiated rutin using the mushroom tyrosinase inhibition method. The natural flavonoid glucoside rutin was easily degraded by gamma-irradiation into three new products: radioruinol (1), isoradiorutinol (2), and radiorutindiol (3). Structures of the new rutin derivatives, including absolute configurations, were elucidated based on spectroscopic interpretation (nuclear magnetic resonance, UV, circular dichroism, [α]D, and fast atom bombardment-mass spectrometry). The tyrosinase inhibitory activities of the newly formed rutin derivatives were evaluated. The new dihydroflavonol derivative radiorutindiol (3), containing two hydroxymethyl groups instead of olefin and ketone moieties, exhibited significantly improved inhibitory activity against tyrosinase with an IC50 value of 45.3±1.6 μM compared to the parent rutin.
