Summary:Objectives : Allergic rhinitis is an IgE-mediated inflammatory disease of the nasal mucosa characterized by sneezing, nasal congestion, and runny nose. Particulate matter (PM) enters the respiratory tract and adheres to the nasal mucosa, amplifying the allergic inflammatory response. Currently, corticosteroids and antihistamines are used to relieve the symptoms of allergic rhinitis, but long-term use can cause serious side effects. In particular, there is a lack of research on the efficacy of natural product-based treatments for allergic rhinitis exacerbated by PM. Therefore, this study aimed to analyze the allergy suppressive (anti-inflammatory) effects of Zingiber officinale Roscoe ethanolic extract (ZR) on a PM-exacerbated allergic rhinitis model. Methods : To evaluate the effect of ZR in a PM-exacerbated rhinitis model, 7-week-old male mice were sensitized with aluminum hydroxide and ovalbumin (OVA), then challenged with nasal instillations of OVA and PM. ZR was orally administered for 13 days, and the mice were sacrificed on day 28. Thereafter, serum, nasal lavage fluid (NALF), and nasal tissue were collected for analysis of relevant inflammatory markers and subjected to histologic and immunohistochemical analysis. Results : In a PM-exacerbated allergic rhinitis model, ZR inhibited the secretion of immunoglobulin E (IgE) in serum and reduced the levels of inflammatory cytokines in NALF. Furthermore, histological and immunohistochemical analyses confirmed that ZR reduced the infiltration of immune-related cells induced by PM. Conclusions : These results suggest that ZR may be a potential therapeutic agent for modulating excessive immune responses in models of allergic rhinitis exacerbated by PM.
