Summary: | The biological characterization and the property of 4 Human Immunodeficiency Virus(HIV) isolates from HIV infected persons in Korea were analyzed and compared with reference HIV strain. They were designated as NIH 1, NIH 2, NIH 3, and NIH 4. When the reference HIV strain and isolates were inoculated into MT-2 cells, multinucleated giant cells were observed around 48 hours after infection. As the results of immunofluorescence assay in MT-2 cells, the viral antigen was detected at 18 hours postinfection in the reference HIV strain, at 24 hours in NIH 2 and NIH 3, and at 48 hours in NIH 1 and NIH 4. When the rate of HIV replication and cell lysis were exarnined in the MT-4 cells using the MTT assay, the reference HIV strain was more rapid than 4 isolates. Among 4 isolates, the NIH 1 replicated rapidly and others didnt show any difference. In the measurement of p24 antigen using HIV-1 p24 antigen capture ELISA, the amount of p24 antigen produced by NIH 1 in the MT-2 cells was calculated as about 670pg/ml, NIH 3 was about 640pg/ml, reference HIV strain and NIH 2 were about 600pg/ml, and NIH 4 was about 340pg/ml. When using the MT-4 cells, that of NIH 1 was about 1,290pg/ml, NIH 3 was about 1,260pg/ml, NIH 2 was about 730pg/ml, and reference HIV strain and NIH 4 were about 680pg/ml. For the production of p24 antigen by the reference HIV strain, the largeamount of p24 antigen was produced early and was maintained afterward continously because of cell lysis. In the MT-2 cells, reverse transcriptase activity of NIH 1, NIH 3 and NIH 4, and NIH 2 were about 6 x 10' cpm, 4 x 10'cpm, and 3 x 10'cpm respectively. The order of which reverse transcriptase activity reached the peak after culture was reference HIV strain, NIH 4, NIH 3, NIH 2, and NIH 1. In conclusion, the infectivity, replication rate, and cell lysis of reference HIV strain were more rapid than those of isolates. And the viral productivity of NIH 1 and NIH 3 was higher than reference HIV strain and other isolates. |
