Abstract | Objectives : Atopic dermatitis (AD) is one of the most common inflammatory skin diseases characterized by defects in the epidermal barrier and immune system dysregulation, manifested by hives, dry skin, and severe eczematous lesions. Sinapine, an alkaloidal amine found in Cruciferae seeds, exhibits various pharmacological effects, including anti-inflammatory and antioxidant properties. However, its effects on AD remain unclear. Therefore, this study aimed to investigate the efficacy and mechanisms of Sinapine on AD.
Methods : To analyze inflammatory cytokine levels and the MAPK signaling pathways, HaCaT cells were pretreated with sinapine and then stimulated with TNF-α/IFN-γ in vitro . For in vivo analysis, BALB/c mice were treated with repeated DNCB and DFE applications to the ears, followed by 4 weeks of oral Sinapine administration. AD severity was assessed using the SCORAD index, scratching behavior, and lesion swelling, while serum IgE, tissue thickness, and inflammatory cell infiltration were evaluated.
Results : In TNF-α/IFN-γ-treated HaCaT cell model, Sinapine significantly reduced the levels of MDC, TARC, IL-6, GM-CSF, IL-1β, and IL-8. It also decreased p-ERK levels and increased p-JNK levels. In DNCB/DFE-applied mouse model, Sinapine significantly inhibited ear and epidermal thickness and the infiltration of mast cells, eosinophils, and CD4+ T cells.
Conclusions : Sinapine modulated ERK and JNK signaling and reduced inflammatory cytokine expression, thereby attenuating inflammatory responses and pathological features associated with AD. These findings suggest that Sinapine may serve as a potential alternative treatment for AD. |
