无刚毛鳞甲复合体同源物样蛋白2在慢性丙型肝炎患者中的表达及其与血清球蛋白的关系

Summary: | 目的:探讨慢性丙型肝炎(CHC)患者和健康志愿者外周血 CD4+CXCR5+PD -1+T 淋巴细胞与血清球蛋白之间的相关性,明确外周血滤泡辅助性 T 淋巴细胞(Tfh)转录因子无刚毛鳞甲复合体同源物样蛋白(Ascl)2的表达在慢性 HCV 感染过程中的作用。方法选取2015年10月-2016年5月第四军医大学唐都医院收治的住院或门诊初治 CHC 患者46例,另选取实验室健康成年实验人员以及本院健康体检者32例作为健康对照组。应用流式细胞技术分析外周血中 Tfh 细胞在 CD4+T 淋巴细胞中所占比例及其亚群(CD4+CXCR5+PD -1+T 淋巴细胞)变化,应用全自动生化分析仪检测血清球蛋白水平,并通过 real -time PCR 检测Tfh 细胞中 Ascl2 mRNA 表达情况;分析各指标变化与 HCV 感染的相关性。计量资料组间比较采用独立样本 t 检验,相关性分析采用 Pearson 相关分析。结果 CHC 患者外周血 Tfh 细胞与 B 淋巴细胞呈正相关(r =0.5823,P =0.0112),外周血 B 淋巴细胞与球蛋白呈正相关(r =0.4509,P =0.0316),Tfh 细胞与球蛋白呈正相关(r =0.5835,P =0.0383);CHC 患者外周血 Tfh 细胞中 Ascl2 mRNA 表达显著高于健康对照组(1.019±0.666 vs 6.437±5.776,t =4.552,P =0.0019)。结论 Tfh 细胞与 CHC 患者血清中球蛋白的产生有一定的联系,转录因子 Ascl2可能参与 HCV 感染过程中 Tfh 细胞的分化发育。 |
