农杆菌介导EPSPS抗草甘膦基因转化马铃薯品种
摘要:以马铃薯品种费乌瑞它微型薯为外植体,利用农杆菌介导法,将EPSPS抗草甘膦基因转入马铃薯,并对其遗传转化的影响因素进行研究。经PPT筛选获得抗性植株17株;经PCR检测,17株呈阳性;RT—PCR检测,6株呈阳性,说明肼峪P5基因能够正常转录;Southernblot杂交分析,5株呈阳性,证明E朋PS基因已整合到马铃薯基因组中。对转基因植株后代进行抗草甘膦试验,转基因材料对浓度0.4g·L-1的草甘膦表现出抗性。...

Bibliography: | 23-1391/S In this study, EPSPS gene was transferred into the potato (Solanum tuberosum L.) cultivars, 'Favorite' by Agrobacterium-mediated transformation, and influence factors of genetic trans- formation were optimized. Microtuber discs were used as explants. Resistant plants were analyzed by PCR detection, of which 17 plants were positive. By RT-PCR test, six strains of 'Favorite' were positive; EPSPS gene could be transcribed nomally. And these resistant plants were analyzed by Southern blot, of which five plants had a positive amplification. Southern blot showed that single copies of the EPSPS gene were inserted into the genome of the transgenic plants.
