解磷芽孢杆菌YC4液体培养基及发酵条件的优化

对从养殖池塘中筛选出的一株具有解磷特性的芽孢杆菌YC4进行液体培养基和发酵条件的优化实验.在摇床培养条件下,研究了各营养源和主要影响因子对菌株YC4生长和芽孢形成的影响,并建立了菌株的生长曲线.结果显示:解磷芽孢杆菌YC4最佳发酵培养基配方为糖蜜10‰、大豆蛋白粉5‰、酵母膏5‰、磷酸二氢钾5‰、硫酸镁1.5‰、氯化钠15‰、玉米浆5‰;最适发酵条件为温度30℃,初始pH值为7.0~7.5,接种量1%,摇床转速200r/min;最适收获时间为24 h.优化后总菌量达1.23×109 CFU/mL,芽孢产量达1.01×109 CFU/mL,明显高于基础培养基发酵结果....

To optimize the liquid fermentation medium and conditions for a phosphate-decomposing Bacillus strain YC4 isolated from aquaculture ponds, nutrient sources and major influencing factors were experimented under shake flasks to obtain their effects on the cell growth and spore formation and the growth curve of the strain was established. The optimal medium is determined to be composed of 10‰ of molasses, 5‰ of soybean protein powder, 5‰ of yeast extract, 5‰ of KH2PO4, 1.5‰ of MgSO4,15‰ of NaCl and 5‰ of corn steep liquor, and the optimal fermentation condition is 30℃ with initial medimn pH of 7.0
