轮枝镰孢菌的绿色荧光蛋白基因标记

本研究采用农杆菌介导法(ATMT)将绿色荧光蛋白基因(gfp)转入轮枝镰孢菌(Fusarium verticillioides)获得了表达gfp的转化子。结果表明,转化子经过5代培养仍能发出稳定的荧光,转化后的菌落形态与菌丝生长速度差异不明显,对寄主仍有致病能力。通过PCR验证也表明gfp基因已成功转入到菌株PX1015基因组中,轮枝镰孢菌绿色荧光蛋白基因转化体的成功构建为该病菌侵染机制的研究奠定了基础。...

In this study,transformants of expressing green fluorescent protein was obtained by delivering gfp gene into Fusarium verticillioides via agrobacterium tumefaciens-mediated transformation( ATMT). The result showed that the transformants had genetic stability and excellent fluorescence after 5 generations,and there were no significant differences in their colony morphology and mycelium growth rate,whose pathogenicity of host infection remained. PCR detection also indicated that the gfp gene had been transferred to the strain F. verticillioides PX1015.
