草菇核不对称分裂与菌种退化的相关性

【目的】探讨草菇细胞核不对称分裂与菌种退化的相关性,揭示草菇菌种退化机理。【方法】根据草菇菌落形态观察并结合RAPD技术筛选出退化的草菇菌株,通过细胞核DAPI染色,观察其细胞核大小、数目,比较正常菌株与退化菌株细胞核的异同。【结果】草菇菌丝细胞为多核,在有丝分裂过程中存在核不对称分裂。退化菌株细胞中形成染色体的数量不同,同一个细胞中可观察到大小不一的细胞核;正常菌株PY与3个退化菌株PYb5、PYb6、PYb7平均每个菌丝细胞的细胞核数目分别为12.78、10.06、8.88和7.26个,不对称分裂核分别占总核数的2.5%、10.5%、15.3%和23.7%。【结论】草菇细胞核不对称分裂可能...

[Objective]The relativity between asymmetric nuclear division and strain degeneration was discussed to elucidate mechanism of strain degeneration in Volvariella volvacea. 【 Method 】 Three degenerated Volvariella volvacea strains were selected according to RAPD markers and colony morphology. The number and size of nucleus of a normal strain and three degenerated strains of Volvariella volvacea, PYb5, PYb6, PYb7, were observed and compared by nucleus staining. 【Result 】The results showed that many nucleuses exited in Volvariella volvacea hyphal cells. Bigger nucleus and smaller nucleus were produced by asymmetric division during mitosis in Volvariella volvacea hyphal cells.Compared with normal strain,the average number of nucleus per cell was less a
