桂野生山金柑的倍性分析和SSR分子鉴定
【目的】研究‘桂野生山金柑’的遗传学特性,为柑橘种质资源的鉴定及创新利用提供理论依据。【方法】采用倍性分析和SSR标记相结合,对‘桂野生山金柑’进行遗传鉴定分析。【结果】倍性分析结果表明‘桂野生山金柑’是二倍体;从24对SSR引物中筛选出6对重复性好、条带清晰、多态性强的核基因组引物,在供试材料中获得等位基因条带共30条,其中具有多样性差异条带25条,多态性比例为79.17%。聚类分析的结果表明不同供试材料的遗传相似系数在0.600-0.802。【结论】‘桂野生山金柑’具有遗传特异性,不同于其他地区起源的山金柑或金柑,是一种新的柑橘种质资源。...

Abstract_FL | [Objective] In order to provide a theoretical basis for the identification and utilization of Gui Wild Shanjingan,its genetic characteristics was studied [Methods] Using ploidy analysis and SSR molecular marker to analyze the genetic diversity of Gui Wild Shanjingan.[Results] The ploidy analysis result showed that Gui Wild Shanjingan was diploid.6 pairs of SSR primers with good repeatability,high polymorphism and clear bands were screened from the 24 pairs of SSR primers.A total of 30 allele stripes were successfully observed,among them,25 bands were polymorphism.The polymorphic rate was 79.17 %.Genetic similarities caculated by cluster analysis ranged from 0.600-0.802.[Conclusion] Gui Wild Shanjingan was significant different from Shanjingan or Jingan originated from other origions,it was a new cultivar with genetic specificity. |
