玉玲花虫瘿中单宁含量初探

首先通过正交实验设计,采用了水解法、酸提取法、乙醇提取法3种不同的提取方法对虫瘿中的单宁进行提取,然后用紫外分光光度法对虫瘿中的单宁进行测定,得出采用乙醇提取法,且在提取温度80℃、料液比1∶30、乙醇浓度60%、提取时间3h的条件下,测得单宁最高含量为10.9%。在此基础上,采用乙醇提取法分别提取玉玲花虫瘿、叶、枝条中的单宁,分别用紫外分光光度法和高效液相色谱法测定单宁含量。结果表明:不同采集时期,虫瘿的含量不同,绿色期为10.9%,失绿期4.7%。紫外分光光度法测得单宁含量叶〉单宁含量虫瘿〉单宁含量枝条,分别为11.9%、4.7%、3.7%,不同部位差异较大;高效液相色谱法测得单宁含量虫瘿...

Notes Tannin in insect gall in Styrax obassia was extracted by methods of hydrolysis,acid extraction and ethanol extraction.The orthogonal experiment results showed that ethanol extraction was most suitable to do the work under a condition of 80℃ temperature,1 ∶30 solid-liquid ratio,60% ethanol concentration and 3 h extraction time.The tannin in the galls was determined by UV spectrophotometry and HPLC.It was found that the content of tannin at green period and chlorina period was 10.9 % and 4.9%,respectively.The tannin content in galls,leaves and stems tested by UV spectrophotometry was 11.9 %,4.7 % and 3.7 %,respectively while the content tested by HPLC was 2.4 %,2.4 % and 2.0 %,respectively.
