旱半夏组织培养技术的优化研究
以旱半夏无菌苗的叶片、叶柄和块茎为外植体,在不同温度及激素组合的条件下,进行了愈伤组织诱导、芽分化和生根的研究。结果表明:愈伤组织诱导的最适温度为25℃;MS+KT 1.0 mg/L+2,4-D 2.0 mg/L对块茎的愈伤诱导效果最好,诱导率达96.7%。温度对芽分化的影响不明显;最佳的芽分化培养基为MS+KT 3.0 mg/L+2,4-D 1.0 mg/L。MS+IBA 0.5 mg/L+蔗糖30 g/L对单芽增殖效果明显。MS+KT 1.0 mg/L+NAA 0.5 mg/L最有利于生根。...

Notes | 36-1124/S Using the leaf,leafstalk and rhizome of the aseptic seedlings of Pinellia ternate Thunb.Breit as explants,the author studied the effects of temperature and plant hormones on the inducement of callus,bud differentiation and rooting.The results showed that the optimum temperature for callus inducement was 25 ℃.The optimum medium for the inducement of callus from rhizome was MS+ KT 1.0 mg/L+ 2,4-D 2.0 mg/L,and the inducement rate reached 96.7%.Temperature had little effect on the bud differentiation.The optimum medium for bud differentiation was MS+ KT 3.0 mg/L+ 2,4-D 1.0 mg/L.The medium MS+ IBA 0.5 mg/L+ cane sugar 30 g/L had significant effect on the proliferation of single bud.The medium MS+ KT 1.0 mg/L+ NAA 0.5 mg/L was most beneficial for rooting.
