紫薇属植物染色体制片技术优化
以紫薇属紫薇(Lagerstroemia indica)、屋久岛紫薇(L.fauriei)、福建紫薇(L.limii)为研究对象,从取材、预处理、制片方法等方面比较了常规压片和去壁低渗法两种染色体制片技术的效果,并进行染色体计数。结果表明:利用新萌发的茎尖在改良固定液(无水乙醇∶冰醋酸∶三氯甲烷=5∶3∶2)中-20℃固定过夜后的制片效果最好;常规压片法适用于普通计数、观察,利用火焰干燥法可获得维持染色体原有形态、能用于荧光原位杂交实验的染色体制片。此外,本研究观察到紫薇、屋久岛紫薇和福建紫薇的染色体数目均为2n=48,福建紫薇和屋久岛紫薇的染色体数目均为初次报道。...

Notes | 44-1267/S Lagerstroemia; chromosome; slide-preparing of chromosomes YANG Bing-jie,CHEN Jing-xin,TANG Wan,WANG Xiao-jiao,LIU Yang,PAN Hui-tang,(1.College of Landscape Architecture,Beijing Foresty University,Beijing 100083,China; 2.National Engineering Research Center for Floriculture,Beijing 100083,China) The effects of different plant materials,pretreatment methods on chromosome sectioning of Lagerstroemia indica,L.limii and L.Fauriei were investigated.The results showed that new stem tip fixed with improved Carnoy solution(EtOH∶HAC∶THMs=5∶3∶2) at-20℃ overnight performed best in the sectioning.The conventional squash method was appropriate for chromosome counting and observation.The enzyme hydrolysis method could maintain the original form of chromosomes and the sections could be used for FISH.The chromosome number of L.indica,L.limii and L.fauriei were all 2n=48,and it was the first time to report the chromosome number of L.limii and L.fauriei. |
