Abstract Paeonia emodi (peony) is a well known plant used medicinally to treat hypertension, palpitations, and asthma. Despite its popularity, there are few reports in the scientific literature examining its use in such conditions. We prepared a 70% ethanolic extract of peony root (Pe.Cr) and applied it to segments of guinea pig atria and trachea and rat aorta suspended separately in tissue baths. Activity against arachidonic acid (AA)-induced platelet aggregation was measured in human platelet-rich plasma. Airway relaxant effect was evaluated against acetylcholine (ACh)-induced airway contraction in mouse lung slices loaded with fluo-4. Pe.Cr (0.3-10 mg/mL) showed an atropine-resistant negative inotropic effect in atria. In rat aorta, an endothelium-independent vasodilatory effect (0.3-10 mg/mL) was seen in phenylephrine- and high-K + -induced contractions. Pe.Cr (0.01-1 mg/mL) also inhibited AA-induced platelet aggregation. In isolated trachea, Pe.Cr (0.3-10 mg/mL) relaxed carbachol- and histamine-induced contractions independently of β-adrenergic receptors. In mouse lung slices, Pe.Cr (0.3-1 mg/mL) inhibited ACh-induced airway narrowing and oscillations of intracellular Ca 2+ in airway smooth muscle cells. The results showed cardiosuppressant, vasodilatory, antiplatelet, and tracheal and airway relaxant activities of peony, providing potential justification for its medicinal use in different hyperactive cardiovascular and respiratory disorders.

Abstract_FL Paeonia emodi, aussi appelée pivoine de l'Himalaya, est une plante médicinale bien connue utilisée pour traiter l'hypertension, les palpitations et l'asthme. Malgré sa popularité, il existe peu d'articles justifiant son utilisation dans de telles situations pathologiques, d'où l'intérêt de la présente étude. On a préparé un extrait éthanolique à 70 % (Pe.Cr) de racines de pivoine. On a suspendu séparément des segments d'oreillettes et de trachées de cobayes et des segments d'aortes de rats dans des bains de tissus. On a mesuré l'activité contre l'agrégation induite par l'acide arachidonique (AA) dans du plasma riche en plaquettes humaines. On a évalué l'effet relaxant sur la contraction bronchique induite par l'acétylcholine (ACh) dans des tranches de poumons murins chargées avec du fluo-4. Pe.Cr (0,3-10 mg/mL) a montré un effet inotrope négatif résistant à l'atropine dans les oreillettes. Dans l'aorte, un effet vasodilatateur indépendant de l'endothélium (0,3-10 mg/mL) a été observé sur les contractions induites par la phényléphrine et par un haut taux de K + . Pe.Cr (0,01-1 mg/mL) a aussi inhibé l'agrégation des plaquettes induite par l'AA. Dans la trachée isolée, Pe.Cr (0,3-10 mg/mL) a relaxé les contractions induites par le carbachol et l'histamine, indépendamment des récepteurs β-adrénergiques. Dans les tranches de poumons de souris, Pe.Cr (0,3-1 mg/mL) a inhibé le rétrécissement des voies aériennes induit par l'ACh et les oscillations du Ca 2+ dans les cellules musculaires lisses bronchiques. Ces résultats montrent que la pivoine a des activités cardio-inhibitrices, vasodilatatrices, antiplaquettaires et relaxantes dans la trachée et les voies aériennes. Ces observations justifient le potentiel de cette plante médicinale pour traiter différents troubles respiratoires et cardiovasculaires.
