Abstract The purpose of this study was to examine the relative proportions of subgingival microbial flora in periodontally diseased patients before and after initial preparation. Plaque control, subgingival scaling and root planing were carried out in nine subjects with adult periodontitis ranged from 4 to 8mm pockets. The bacterial samples were obtained and subjected to Gram stain, dark field microscopy and an anaerobic incubation prior to treatment, after plaque control, and after scaling and root planing. The relative number of Gram positive cocci increased while Gram negative rods and filaments decreased after initial treatment. The mean percentage of Fusobacterium was 6.8%, 4.8% and 1.4%. Bacteroides was 11.0%, 3.1% and 0.1%, Spirochetes was 11.0%, 9.5% and 0.5%, respectively, black-pigmented Bacteroides and Fusobacterium necrophorum could be characterized and identified by the analyses of the gas chromatography and “Anaerobic Laboratory Manual”. A positive correlation was observed between Bacteroides and probing depth. Spirochetes closely correlated with probing depth, gingival index and plaque score as clinical parameters.
